    Exposure-Tolerant Imaging Solution forCultural Heritage Monitoring

    This paper describes a simple and cheap solution specifically designed for monitoring the degradation of thin coatings employed for metal protection. The proposed solution employs a commercial photocamera and a frequency-domain-based approach that is capable of highlighting the surface uniformity changes due to initial corrosion. Even though the proposed solution is specifically designed to monitor the long-time performance of protective coatings employed for the restoration of silver artifacts, it can be successfully used also for assessing the conservation state of other ancient metallic works of art. The proposed solution is made tolerant to exposure changes by using a procedure for sensor nonlinearity identification and correction, does not require a precise lighting control, and employs only free open-source software, so that its overall cost is very low and can be used also by not specifically trained operator

    Health-related quality of life in patients with Gilles de la Tourette syndrome at the transition between adolescence and adulthood

    Gilles de la Tourette syndrome (GTS) is a neurodevelopmental condition characterised by tics and comorbid behavioural problems, affecting predominantly male patients. Tic severity typically fluctuates over time, with a consistent pattern showing improvement after adolescence in a considerable proportion of patients. Both tics and behavioural co-morbidities have been shown to have the potential to affect patients’ health-related quality of life (HR-QoL) in children and adults with persisting symptoms. In this study, we present the results of the first investigation of HR-QoL in patients with Gilles de la Tourette syndrome at the transition between adolescence and adulthood using a disease-specific HR-QoL measure, the Gilles de la Tourette Syndrome-Quality of Life-Children and Adolescents scale. Our results showed that patients with GTS and more severe co-morbid anxiety symptoms reported lower HR-QoL across all domains, highlighting the impact of anxiety on patient’s well-being at a critical stage of development. Routine screening for anxiety symptoms is recommended in all patients with GTS seen at transition clinics from paediatric to adult care, to implement effective behavioural and pharmacological interventions as appropriate

    In this study the use of green tea extract (GTE) for the production of frozen carrots fortified with GT polyphenols was investigated. To this purpose, sliced carrots (0.5 cm) were pre-treated by blanching (90 °C; 180 sec) and vacuum impregnated (50 mbar; 10 minutes) in water or GT extract solution (0.25% w/w), and thereafter frozen (-40 °C) and stored at -18 °C for 60 days. In order to evaluate the effect of processing on the functional properties, the overall quality and stability of carrots, fresh and pre-treated samples were evaluated before and after frozen storage for their colour, firmness, total carotenoids and polyphenols content (TPC), and antioxidant activity (AOA)

    Use of Conventional and Innovative Technologies for the Production of Food Grade Hop Extracts: Focus on Bioactive Compounds and Antioxidant Activity

    This study investigated the use of conventional and innovative extraction methods to produce food-grade hop extracts with high antioxidant capacity and content in bioactive compounds. Conventional extractions (CONV) were performed under dynamic maceration at 25 and 60 °C; innovative extractions were performed using two ultrasound systems, a laboratory bath (US) and a high-power ultrasound bath (HPUS), and a high-pressure industrial process. For CONV, US, and HPUS extractions the effect of the extraction time was also tested. Experimental results showed that extraction method, temperature, and time affect to a different extent the phenolic profile and have a significant effect (p < 0.05) on the total phenolic content, total flavonoid content, antiradical capacity (ABTS), chlorophyll α, and total carotenoids content. Overall, US and CONV 60 °C extractions showed the highest extraction efficiency for almost all the investigated compounds, however, the extraction method and time to be used strongly depends on the target compounds to extract

    Lipid peroxidation and apoptotic response in rat brain areas induced by long-term administration of nandrolone: the mutual crosstalk between ROS and NF-kB

    The aim of this study was to evaluate the played by oxidative stress in the apoptotic response in different brain areas of rats chronically treated with supra-physiological doses of nandrolone decanoate (ND). Immunohistochemical study and Western blot analysis were performed to evaluate cells' apoptosis and to measure the effects of expression of specific mediators, such as NF-ÎșB (nuclear factor kappa-light-chain-enhancer of activated B cells), Bcl-2 (B-cell lymphoma 2), SMAC/DIABLO (second mitochondria-derived activator of caspases/direct IAP-binding protein with low PI) and VMAT2 (vesicular monoamine transporter 2) on apoptosis. The results of the present study indicate that a long-term administration of ND promotes oxidative injury in rat brain specific areas. A link between oxidative stress and NF-ÎșB signalling pathways is supported by our results. In addition to high levels of oxidative stress, we consistently observed a strong immunopositivity to NF-ÎșB. It has been argued that one of the pathways leading to the activation of NF-ÎșB could be under reactive oxygen species (ROS)-mediated control. In fact, growing evidence suggests that although in limited doses, endogenous ROS may play an activating role in NF-ÎșB signalling, while above a certain threshold, they may negatively impact upon this signalling. However, a mutual crosstalk between ROS and NF-ÎșB exists and recent studies have shown that ROS activity is subject to negative feedback regulation by NF-ÎșB, and that this negative regulation of ROS is the means through which NF-ÎșB counters programmed cells

    Abstract Vacuum Impregnation (VI) is a non-destructive technology, used to introduce external liquids, in the porous structures of food matrices favored by the action of hydrodynamic mechanism as promoted by pressure changes. Previous studies evidenced the potentiality to apply VI to zucchini [2] but no systematic studies have been carried out yet. Aim of this study was to investigate the effect of VI by using mixed solutions containing structuring ability components (maltodextrins, salt and CaCl2) with on quality and microstructural properties of zucchini. Slices of zucchini (0.5-cm thick) ( Cucurbita pepo, L.) were subjected to VI treatments. VI solutions with different solutes composition were investigated: maltodextrines, NaCl and CaCl2 were used to prepare single (1 solute) or mixed (2 and 3 solutes) VI solutions Samples were analysed for total solids, soluble solids, salt and [Ca] content, textural properties (shear force, relaxation test), sensory and microstructure (Cryo-SEM). Mass balance (Solute Gain/Loss, Water Gain/Loss) was also computed. The composition of the VI solution affected mass transfer. A significant change in the textural properties was observed in zucchini processed with VI solutions containing NaCl and MD; the presence of CaCl2 in the VI solution in combination with the other solutes, was able limit the hardness loss and when alone even to determine an hardening effect. Cryo-SEM analysis highlighted a different effect on microstructure of the vegetable tissue due the use of the single or mixed VI solutions. The use of VI mixed solutions made with MD, NaCl and CaCl2 to process zucchini allows to favour solute and water gain while limiting textural and microstructural change

    Nuclear Diacylglycerol Produced by Phosphoinositide-specific Phospholipase C Is Responsible for Nuclear Translocation of Protein Kinase C-α

    It is well established that an independent inositide cycle is present within the nucleus, where it is involved in the control of cell proliferation and differentiation. Previous results have shown that when Swiss 3T3 cells are treated with insulin-like growth factor-I (IGF-I) a rapid and sustained increase in mass of diacylglycerol (DAG) occurs within the nuclei, accompanied by a decrease in the levels of both phosphatidylinositol 4-phosphate and phosphatidylinositol 4,5-bisphosphate. However, it is unclear whether or not other lipids could contribute to this prolonged rise in DAG levels. We now report that the IGF-I-dependent increase in nuclear DAG production can be inhibited by the specific phosphatidylinositol phospholipase C inhibitor 1-O-octadeyl-2-O-methyl-sn-glycero-3-phosphocholine or by neomycin sulfate but not by the purported phosphatidylcholine-phospholipase C specific inhibitor D609 or by inhibitors of phospholipase D-mediated DAG generation. Treatment of cells with 1-O-octadeyl-2-O-methyl-sn-glycero-3-phosphocholine or neomycin sulfate inhibited translocation of protein kinase C-alpha to the nucleus. Moreover, exposure of cells to 1-O-octadeyl-2-O-methyl-sn-glycero-3-phosphocholine, but not to D609, dramatically reduced the number of cells entering S-phase upon stimulation with IGF-I. These results suggest that the only phospholipase responsible for generation of nuclear DAG after IGF-I stimulation of 3T3 cells is PI-PLC. When this activity is inhibited, neither DAG rise is seen nor PKC-alpha translocation to the nucleus occurs. Furthermore, this PI-PLC activity appears to be essential for the G0/G1 to S-phase transition

    MiR-23-TrxR1 as a novel molecular axis in skeletal muscle differentiation

    Thioredoxin reductase 1 (TrxR1) is a selenocysteine-containing protein involved in cellular redox homeostasis which is downregulated in skeletal muscle differentiation. Here we show that TrxR1 decrease occurring during myogenesis is functionally involved in the coordination of this cellular process. Indeed, TrxR1 depletion reduces myoblasts growth by inducing an early myogenesis -related gene expression pattern which includes myogenin and Myf5 up-regulation and Cyclin D1 decrease. On the contrary, the overexpression of TrxR1 during differentiation delays myogenic process, by negatively affecting the expression of Myogenin and MyHC. Moreover, we found that miR-23a and miR-23b - whose expression was increased in the early stage of C2C12 differentiation - are involved in the regulation of TrxR1 expression through their direct binding to the 3â€Č UTR of TrxR1 mRNA. Interestingly, the forced inhibition of miR-23a and miR-23b during C2C12 differentiation partially rescues TrxR1 levels and delays the expression of myogenic markers, suggesting the involvement of miR-23 in myogenesis via TrxR1 repression. Taken together, our results depict for the first time a novel molecular axis, which functionally acts in skeletal muscle differentiation through the modulation of TrxR1 by miR-23

    Abstract Introduction In Tuscany, Italy, where a universal immunization program with monovalent meningococcal C conjugate vaccine (MCC) was introduced in 2005, an outbreak of invasive meningococcal disease (IMD) due to the hypervirulent strain of Neisseria meningitidis C/cc11 occurred in 2015–2016, leading to an immunization reactive campaign using either the tetravalent (ACWY) meningococcal conjugate or the MCC vaccine. During the outbreak, IMD serogroup C (MenC) cases were also reported among vaccinated individuals. This study aimed to characterize meningococcal C conjugate vaccines (MenC-vaccines) failures and to estimate their effectiveness since the introduction (2005–2016) and during the outbreak (2015–2016). Methods MenC cases and related vaccine-failures were drawn from the National Surveillance System of Invasive Bacterial Disease (IBD) for the period 2006–2016. A retrospective cohort-study, including the Tuscany' population of the birth-cohorts 1994–2014, was carried out. Based on annual reports of vaccination, person-years of MenC-vaccines exposed and unexposed individuals were calculated by calendar-year, birth-cohort, and local health unit. Adjusted (by birth-cohort, local health unit, and calendar-year) risk-ratios (ARR) of MenC invasive disease for vaccinated vs unvaccinated were estimated by the Poisson model. Vaccine-effectiveness (VE) was estimated as: VE = 1-ARR. Results In the period 2006–2016, 85 MenC-invasive disease cases were reported; 61 (71.8%) from 2015 to 2016. Twelve vaccine failures occurred, all of them during the outbreak. The time-interval from immunization to IMD onset was 20 days in one case, from 9 months to 3 years in six cases, and ≄7 years in five cases. VE was, 100% (95%CI not estimable, p = 0.03) before the outbreak (2006–2014) and 77% (95%CI 36–92, p  Conclusions In Tuscany, MenC-vaccine failures occurred exclusively during the 2015–2016 outbreak. Most of them occurred several years after vaccination. VE during the outbreak-period was rather high supporting an effective protection induced by MenC-vaccines
